چکیده

This chapter is a review of the evidence implicating a dysfunction of dopamine transmission inthe prefrontal cortex in schizophrenia, underlying negative symptoms and cognitive deficits. The evidencederives essentially from brain imaging studies, post-mortem studies and clinical pharmacological studies. The imaging studies have suggested alterations in the main mediator of dopamine transmission in thecortex, the D1 receptors. Furthermore, studies have suggested a link between NMDA deficits and D1alterations in the cortex in schizophrenia, suggesting that therapeutic interventions at these targets mayhave beneficial effects on cognition and negative symptoms. List of Abbreviations: CKU, chronic ketamine users; COMT, catechol-O-methyltransferase; DA, dopa-mine; DAT, DA transporters; DLPFC, dorsolateral prefrontal cortex; EC, entorhinal cortex; MPFC, medial.1 Cortical dopamine in schizophrenia prefrontal cortex; NET, norepinephrine transporters; PFC, prefrontal cortex; SN, substantia nigra; VTA,ventral tegmental area; TH, tyrosine hydroxylase; WCST, Wisconsin Cart Sort Task; WM, working memory

Cambridge Medical Reviews: Neurobiology and Psychiatry

In the ensuing years there have been many studies searching for the abnormal anatomy and toxic substances. These two lines of study examine complementary issues of brain abnormalities apparent at the levels of structural and neurochemical anatomy. In the study of the post-mortem schizophrenic brain there are many important questions which are being addressed by modern neuropathology and neuroch...

Psychosis as a state of aberrant salience: a framework linking biology, phenomenology, and pharmacology in schizophrenia.

OBJECTIVE The clinical hallmark of schizophrenia is psychosis. The objective of this overview is to link the neurobiology (brain), the phenomenological experience (mind), and pharmacological aspects of psychosis-in-schizophrenia into a unitary framework. METHOD Current ideas regarding the neurobiology and phenomenology of psychosis and schizophrenia, the role of dopamine, and the mechanism of...

Up-regulation of NMDA receptor subunit and post-synaptic density protein expression in the thalamus of elderly patients with schizophrenia.

Numerous studies have described structural and functional abnormalities of the thalamus in schizophrenia, but surprisingly few studies have examined neurochemical abnormalities that accompany these pathological changes. We previously identified abnormalities of multiple molecules associated with glutamatergic neurotransmission, including changes in NMDA receptor subunit transcripts and binding ...

Advances in post mortem molecular neurochemistry and neuropathology: examples from schizophrenia research.

Post mortem studies of psychiatric disorders have been revitalised by several developments. Molecular biology has provided tools for studying genes and their expression in post mortem brain tissue, an approach which facilitates integration of molecular genetics with neurochemistry and neuropathology. The techniques can be used quantitatively as well as qualitatively, applications which have bee...

Sensing the Worst: Neurophenomenological Perspectives on Neutral Stimuli Misperception in Schizophrenia Spectrum

While investigating social cognitive impairments in schizophrenia, prominent evidence has been found that patients with schizophrenia show a tendency to misclassify neutral stimuli as negatively valenced. Within this population, patients presenting delusions are more prone to this phenomenon. In a previous study, Schizophrenia spectrum (SzSp) patients rated positive, negative and neutral stimul...
